Protocol for Pre-cast Gel Protein Electrophoresis Experiment

Experimental Preparation

Check Equipment: Ensure that the protein electrophoresis chamber, power supply, and transfer system are in working order. We offer DYCZ-24DN for protein electrophoresis, DYCZ-40D for transfer system, and DYY-6C for power supply.

Sample Preparation: Prepare your samples according to the experimental design. Treat protein samples with reducing agents and proteases if necessary.

Prepare Electrophoresis Buffer: Follow the instructions provided with the pre-cast gel to prepare the electrophoresis buffer at the appropriate concentration.

Handling the Pre-cast Gel:

Remove Pre-cast Gel: Carefully open the packaging and remove the pre-cast gel from its container, taking care not to damage the gel matrix.

Sample Loading: Load your prepared samples into the sample wells of the gel using a micropipette or other suitable tools. Pay attention to the loading order and the volume of each sample.

Set Electrophoresis Conditions: Set the electrophoresis conditions, including current intensity, voltage, and duration. Ensure you choose conditions suitable for optimal separation.

Running the Electrophoresis

Initiate Electrophoresis: Place the gel in the electrophoresis chamber, connect the power supply, and start the electrophoresis. Monitor the process to ensure a stable current.

Complete Electrophoresis: Stop the electrophoresis when the samples have migrated to the desired positions. Avoid running the electrophoresis for too long to prevent samples from running out of the gel.

Transferring Proteins

Prepare Transfer System: Take the gel plate out of the chamber and prepare for protein transfer. This may involve cutting the membrane and preparing the transfer buffer.

Assemble Transfer Setup: Assemble the protein transfer setup according to the instructions provided with the transfer system. Pay attention to the assembly order and setting conditions.

Run Protein Transfer: Initiate the protein transfer process and monitor its progress. Ensure that the transfer time and conditions meet the requirements of your experiment.

Post-Transfer Processing:

Membrane Handling: Process the transferred membrane as needed, which may include staining, immunoblotting, or other post-transfer procedures based on experimental requirements.

Result Analysis: Analyze the results based on your experimental design and processing steps. Interpret the findings and generate relevant charts or graphs.
