Polyacrylamide Gel Electrophoresis

Polyacrylamide is often used in molecular biology applications as a medium for electrophoresis of proteins and nucleic acids in a technique known as PAGE. It is a kind of zone electrophoresis method by synthetics gel called polyacrylamide as a supporting medium. It was built by S.Raymond and L.Weintraub in 1959, and then promoted and developed by L.Ornstein and B.J. Davis. This method was widely used after further explanation and modification by them in theory and experimental technique in 1964.

Before applying polyacrylamide gel electrophoresis, people mainly use paper electrophoresis for zone EP. But the paper act as a medium, just have the function of anti-convection, no any other positive effect. While the polyacrylamide gel not only has the function of anti-convection but also it can be participated in the separation proceeding actively. Because polyacrylamide gel is a kind of net structure, which is a polymerization and cross-linking combination of acrylamide (Acr) and N,N-methylenebis(acrylamide). Acrylamide is called monomer, while the N,N-methylenebis is called a comonomer or crosslinker. The forming of a gel is a process of chemical polymerization. The pore size of a gel can be controlled, thus the gel can be made with different cross-linking degrees. If the pore size is approaching the average radius of the sample’s molecule, the resistance of the molecule to go through the gel pore will have a close relationship with the size and shape of the molecule during the electrophoresis. So it provides a changeable separation factor for separating those materials with similar net charges.

There are two general ways used for polyacrylamide gel electrophoresis, one is disc electrophoresis, and the other one is slab electrophoresis. The slab electrophoresis is specially designed for separating protein and DNA, and generally, there are two kinds of slab electrohporesis, which are the horizontal electrophoresis tank and the vertical electrophoresis tank. For the protein, people use the horizontal electrophoresis tank for IFF and immunoelectrophoresis, otherwise, people use the vertical electrophoresis tanks for proteins.
