What is Cellulose Acetate Membrane Electrophoresis?

Cellulose Acetate Membrane Electrophoresis is one type of electrophoresis techniques which uses cellulose acetate membrane as a supporting media for experiments.

Cellulose Acetate is a kind of acetate of cellulose which is acetylated from cellulose’s hydroxyl. When it is soluble in organic solution such as acetone, it can be coated to a homogeneous and fine film with micro-pores. The film thickness is around 0.1-0.15mm that is suitable for experiment.

The advantages of cellulose acetate membrane

Comparing with fliter paper, it has the following advantages:

1.Good effect of separation. For the protein samples, cellulose acetate membrane absorbs few of them, after dye, it can completely decolorize from background color. The dye bands are very clear, so it promotes the accuracy for quantitative determin ation.
2.Fast and save time. The cellulose acetate membrane is less  hydyophilic than fliter paper, so the electro-osmosis performance is less than fliter paper, and the electric current is mainly conducted by the samples that can short the separation time and less the electrophoresis time. Normally, the cellulose acetate membrane electrophoresis time is about 45-60minutes. The whole process of electrophoresis including dyeing and decoloration only needs 90 minutes.
3.High sensitivity and less samples to be used. It needs only 2 μL serum in serum protein electrophoresis.
4.Wide application. Some of protein are not easy to separate on paper electrophoresis, like Alpha-fetoprotein, bacteriolytic enzyme, insulin, and histone etc. But it is more better to use cellulose acetate membrane electrophoresis to separate them.
5.Easy to keep and quantification. After dyeing the cellulose acetate membrane electrophoresis samples, immerse the samples in the mixed solution of glacial acetic acid and ethanol to make a transparent dry plate that be saved for a long time.

Because the cellulose acetate membrane electrophoresis is easy to operate, now it is widely used in analysis and testing plasma protein, lipoprotein,  glycoprotein, Alpha-fetoprotein, enzyme, polypeptide, nucleic acid and other biomacromolecule.

Preparation for Cellulose Acetate Membrane Electrophoresis

 The equipment used includes a low voltage power supply, a horizontal electrophoresis tank. The supporting media is cellulose acetate membrane.

 The reagents:

1.pH8.6 barbitol buffer solution:Diethyl barbituric acid, Diethyl sodium pentobarbital;
2.Stain:Ponceau S, Trichloroacetic;
3.TBS/T or PBS/T: 95% ethanol, glacial acetic acid;
4.Cleaning Solution: anhydrous ethanol, glacial acetic acid.

 Notes about experiment:

1.Load the sample on the rough side:1-2UL is suitable
2.Constant current electrophoresis: current strength 0.4~0.6m A/cm
3.pH8.6 barbitol buffer solution:Ionic strength 0.06
4.Dyeing time: 5 minutes enough
5.Preservation: put the dry elecrtophoresis map in cleaning solution for 10-15 minutes, and then take it out and stick it on a clean glass, after it drys, it will become a transparent film map.
